I'm having the same issue but can give a bit more detail.
32-bit 2D version of Ubuntu 11.10 on older laptop (IBM R60 thinkpad) WM is unity 2D
Software center was used (successfully) approx 11/26/11
Became unavailable after software package update ran as of todays date. which, per the history monitor, included updates to the Package manager, unity 2d, unbuntuone, etc
Internet connection is perfectly functional (update, browser, mail, etc all normal)...I also have a desktop running OO 11.10 full instal (Xeon, 4gb, Nvidia) which both updated normally and successfully installed from the software center as of just prior to this addendum.
Repository information, standard keys, checked items are all normal (unmodified by me except via downloaded software/updates) since install. keys are present and are defaults. Software installed includes both GTK and KDE.
Server is US

Part two of the error above occurs after the the connection error. SCenter then displays the following in a message box;
The action would require the installation of packages from not authenticated sources.
...for packages that are authenticated. (package was IDLE Python 3.2)

Restart/reboot does not solve the issue.
