use dd which will perform a bit by bit copy. Assuming the USB is /dev/sdb you can run:

dd if=/dev/sdb of=~/image.iso bs=1024

and the resulting image.iso will be an image of the USB stick including the boot info. Obviously change the /dev/sdb if the USB is not that.

dd: opening `/dev/sdb': Permission denied
and how to know it is sdb

none 497M 420K 496M 1% /run/shm
/dev/sdb1 3.8G 920M 2.9G 24% /media/LIVE

I want to convert this Live UBUNTU 12.04 USB to ISO file

sudo umount /dev/sdb1
sudo dd if=/dev/sdb of=~/image.iso bs=1024

Will make an image of sdb in your home folder.

Thanks Andrew, It did worked Initially I got an image.iso in home folder which size is pretty heavy around 4.1Gb around, and it was locked too. So I followed the steps you gave to unlock it earlier. And used Zsync to test with the previous one.
And this "reading seed file ubuntu-12.04-desktop-i386.iso: ***" process continued for a long time. And in the end got the below result.

Read ubuntu-12.04-desktop-i386.iso. Target 99.9% complete. ********************************************************************************************************************
downloading from
#################### 100.0% 57.1 kBps DONE

verifying download...checksum matches OK
used 734330880 local, fetched 1032771
rohit@rohit-Aspire-4720Z:~$ md5sum ubuntu-12.04-desktop-i386.iso
d791352694374f1c478779f7f4447a3f ubuntu-12.04-desktop-i386.iso

I am happy now, Saving my ISO is my personal folder. Thanks for your help. So Great to know how to do this..

Thanks alot. :)
