You need to run the update with sudo privileges.

sudo apt-get update
sudo apt-get upgrade

Hi,
make sure you're not running more than one update application at the same time.
e.g.
If you run update manager, close Synaptic.
If you run Add and Remove, close Synaptic.

Check in system administration -> system monitor if a process of a closed update application is still running, right click and kill.

If still having trouble updating, close all running update apps and type in a terminal:
sudo dpkg --configure -a
