How to calculate 100 in the above example?

I do not really understand what you mean.

If you write down 100, then it is not calculated, it might be guessed or measured.

if you mean measure: see faq 1686

In some cases Sikuli IDE's Preview might be helpful or a helper script, that reports mouse location via Env.getMouseLocation().
