How to change the icon of an application?

I open files as administrator and navigate to the application. Changing the line "Icon=..." to "Icon=/usr/share/icons/newicon.png" doesn't change the icon. I used Scratch to edit the file, and the file does indeed save.
Any help is appreciated!

I do recommand to add the icon to /usr/share/icons/hicolor/. To apply the theme-change you then have to execute:

sudo gtk-update-icon-cache /usr/share/icons/hicolor
