How to make 'rich black' :50C 50M 50Y 100K?

I want to make a custom color: rich black: C50 M50 Y50 K100.
Every time I enter the values they jump back to C0 M0 Y0 K100
Is there a solution for this problem?

The CMYK selector in Inkscape (if you have selected the tab labelled CMYK) doesn't work like that, no. It's just another color input widget, it does not produce actual CMYK colors, but RGB.

It might be possible if you enable color management.

However, I'd suggest you stick to RGB in Inkscape and use a DTP software like Scribus to do the conversion for you.

Inkscape cannot export any CMYK values anyway, except for use with Scribus, if you save as SVG, and the process is not very reliable, in my experience.
