You can't vary the thickness of a single line, but you can still achieve the effect you're after by turning that line into a closed shape that can be filled with colour.

After creating your C shape, with it still selected, go to Path > Stroke to Path.
Now select the node editing tool (second one down)
You should see little squares along the line; these are the nodes.
Drag the nodes (and/or its handles) to change the curve; this should create the appearance of making the line thicker at, and around, that point.

Optionally, double click anywhere on the line to create a new node at that point, and fine tune the thickness along the length of your curve.

Note: By default the shape is filled. If you instead get an outline of a crescent, open the fill/stroke properties (Object > Fill and Stroke...), click the plain box (Flat colour) under Fill, and pick a colour.
