Is there a preferred format for the backup file format?

What do you mean? Deja Dup uses its own format (or really, the backup tool duplicity uses its own format and Deja Dup uses duplicity).

I've installed Deja Dup and want to setup an external USB hard drive.
What is the preferred format for the partition on the external hdd for
the Deja Dup generated backup files? Fat 32, ext 4, ext 2. I'm using
Linux Mint 11 with ext4 file system.

It doesn't really matter. Deja Dup's backup files can survive in any drive format. I might suggest fat32 just so you can avoid any permission confusion.

didn't understand why, now I do.
